Comparison of Pituitary Response to Regular GnRH, Analogue (D–TRP⁶) and Placebo

Pituitary response to synthetic regular GnRH, to a potent analogue (D‐TRP⁶) and to placebo were compared in ten azoospermic males. FSH and LH were measured prior to and at given intervals following administration of each substance. In addition, plasma levels of testosterone and prolactin were measured. There was no significant difference in the magnitude of FSH and LH release following injection of their the regular or the analogue form of GnRH. However, plasma gonadotrophins remained elevated for significantly longer time periods following the administration of the analogue GnRH. In those patients in whom LH levels remained elevated for at least 24 hours the observation of elevated testosterone levels permitted the inference of adequate biological activity of endogenously produced LH. Patients who did not respond to the regular GnRH were also non‐responsive to D‐TRP⁶ GnRH. A surprising finding was elevated prolactin levels 4–6 hours following GnRH administration. Placebo had no influence on gonadotrophins, testosterone and prolactin. Vergleichende Untersuchung der Ansprechbarkeit der Hypophyse auf reguläres GnRH, Analog (D‐TRP⁶) und Placebo Es wurde die hypophysäre Ansprechbarkeit auf das reguläre synthetische GnRH, ein potentes Analog (D‐TRP⁶) und Placebo bei zehn azoospermen Männern untersucht. Plasmaspiegel von FSH und LH wurden vor und zu festgelegten Zeiten nach Verabreichung der Substanzen gemessen. Zusätzüch wurden Plasmaspiegel von Testosteron und Prolaktin gemessen. Beide GnRH‐Präparationen bewirkten ähnliche Anstiege der Gonadotropinwerte. Allerdings blieben FSH‐ und LH‐Spiegel nach Injektion des Analogs signifikant länger erhöht. Bei denjenigen Patienten, bei welchen LH‐Werte für mindestens 24 Stunden erhöht blieben, gestattete die Beobachtung ansteigender Testosteronspiegel den Rückschluß auf die adäquate biologische Aktivität des endogen produzierten LH. Diejenigen Patienten, welche nach Verabreichung des regulären GnRH nicht mit einem Anstieg der Plasmawerte für FSH und LH reagiert hatten, sprachen auch nicht auf D‐TRP⁶ an. Ein überraschender Befund waren erhöhte Prolaktinwerte 4–6 Stunden nach Verabreichung des GnRH‐Analogs. Placebo beeinflußte weder die Gonadotropinspiegel noch die Testosteron‐ oder Prolaktinwerte. 1980 Blackwell Verlag GmbH
