A Novel Facial Cream Based on Skin-penetrable Fibrillar Collagen Microparticles

Abstract

BACKGROUND:Collagenproteinplaysanotablerolemaintainingfirmskin.Topicalcreamscontainingcollagenfibersarewidelyavailable,buttheir usefulnessisquestionableduetolimitedskinpenetration.Whenappliedinacream,collagendoesnotpenetratetheskinleavingtheskinstructure unaffected.OBJECTIVE:Weformulatedmicronizedcollageninacreambase.Usinghumanskinsamples,wesoughttoinvestigatetheabilityof themicronizedcollagencreamtopenetratehumanskin.METHODS:Particlesizesofmicronizedmarinecollagenwereevaluatedusingelectron microscopy.Opticalprofilometrywasconductedtoevaluateskintopographyandroughness.Theantioxidantactivityofthecollagenwasevaluated usingtheelectronparamagneticresonancetechniquebymeasuringthechangesinfreeradicalproduction.Collagenpenetrationdepthinhuman skinsampleswasmonitoredusinganon-invasiveopticaltechniqueknownasiterativemultiplaneopticalpropertyextraction,whichworksbased onthedetectionoflaserlightphasechangesfollowingthepresenceofcollagenparticlesindeepskinlayers.RESULTS:Accordingtotheelectron microscopy,collagenparticleswerefoundtobeofvarioussizes,thesmallestbeingabout120nmindiameter.Skintopographymeasurements revealedthatthetreatedcollagencreamincreasedskinsmoothnessofthesamples.Ourresultsderivedfromtheiterativemultiplaneopticalproperty extractionindicatedthatmicronizedcollageninacreambasepenetratesboththestratumcorneumandthedeepepidermallayerstowardthe dermis.CONCLUSION:Ourinvestigationsuggeststhatthecollageninthestudiedcreamformulationwasabletopenetratethestratumcoreum and deep epidermal layers in human skin samples.
